Abstract:
To provide a surface plasmon resonance fluorescence spectroscopy that can determine an enhancement angle at which plasmon scattering light is maximum without retreating an excitation light cut filter from an optical path.SOLUTION: A surface plasmon resonance fluorescence spectroscopy includes the steps of: irradiating a metal film with excitation light via a prism as changing an angle of incidence with respect to the metal film, and detecting intensity of plasmon scattering light exiting from on the metal film and reflected by an excitation light reflection filter to determine an enhancement angle; arranging a detected substance marked by a fluorescence substance on the metal film; and irradiating the metal film with the excitation light via the prism so that the angle of incidence with respect to the metal film is maximum, and detecting intensity of fluorescence discharged from the fluorescence substance marking the detected substance and transmitting the excitation light reflection filter. An amount of light of the excitation light in the step of determining the enhancement angle is greater than that in the step of detecting the intensity of fluorescence.SELECTED DRAWING: Figure 2
