Abstract:
PURPOSE: To obtain a sustained release microcapsule high in the content of a water-soluble physiologically active substance and reduced in the initial release and side effects.
CONSTITUTION: The sustained release microcapsule contains an easily soluble amorphous water-soluble physiological substance having a solubility of ≥1g, preferably ≥5g, especially ≥10g, in 100ml of water at 20°C, and a polymer, preferably a biodegradable polymer. The microcapsule is obtained by dispersing the amorphous water-soluble physiologically active substance in the organic solvent solution of the polymer, dispersing the dispersion in a water phase and subsequently subjecting the prepared S/O/W type emulsion to a drying-in- water treatment. The physiologically active substance is especially preferably a peptide compound such as a luteinizing hormone-releasing hormone. The polymer is preferably a polyester, especially a lactic acid-glycolic acid copolymer or hydroxybutyric acid-glycolic acid copolymer.
