Abstract:
PROBLEM TO BE SOLVED: To provide a new method for synthesizing 2-acetyl-2-amino-5,8-dimethoxy-1,2,3,4-tetralin, which is an important intermediate for amrubicin, where the method has gentle process conditions, is environmentally friendly, is easy to operate, and is suitable for industrial production.SOLUTION: A method comprises, for example, preparing a polysubstituted tetralin compound I-1 from a compound II-2 shown in the following formula through two steps, and synthesizing 2-acetyl-2-amino-5,8-dimethoxy-1,2,3,4-tetralin by going through the following step (A) or (B): (A) a step of hydrolyzing an ester moiety in the compound I-1, and then conducting deprotection of carbamate obtained by treatment with azide in the presence of alcohol; and (B) subjecting the ester moiety in the compound I-1 to amidation with ammonia, and then conducting Hofmann degradation.SELECTED DRAWING: None
