PURPOSE: To obtain plant cells adaptable to higher temperatures, by expressing a transformation recognition gene under the control of a soybean heat shock gene promoter to transform plant cells with an Agrobacterium species T-DNA.
CONSTITUTION: A soybean heat shock gene containing a soybean heat shock gene promoter is first isolated, to which a transformation recognition gene attached with a polylinker is linked to prepare a recombinant soybean heat shock gene, which is then cloned into an Agrobacterium species T-DNA shuttle vector to obtain a recombinant DNA fragment with both the above recombinant gene and the vector taken thereinto. Subsequently, the DNA fragment is transformed into a bacterial strain, which, in turn, is mixed with a 2nd bacterial strain bearing a helper plasmid, the resulting mixture is then transferred into a 3rd bacterial strain, followed by selecting a recombinant among endogenous plasmids and then infecting a plant with the above recombinant. Thereafter, plant cells preserving the recombinant soybean heat shock gene are selected, thus obtaining the objective plant adaptable to higher temperatures.
