PURPOSE: To provide a protein fragment which enables the production and accumulation of significant quantities of LDL receptor fragment which has affinity for and recognizes LDL and gene fragment and yet which is soluble in aqueous solutions, and the method for preparing the protein fragment.
CONSTITUTION: The method for preparing the protein fragment, which has affinity for LDL in a heterologous eukaryotic host cell and is soluble in aqueous solution, is comprised of the steps of constructing a nucleotide sequence comprising a protein-coding sequence, coding for a truncated form of a low density lipoprotein receptor gene which does not include the membrane spanning domain and adjoining a regulatory sequence effective to express the protein-coding sequence in the host cell, then transforming the nucleotide sequence thus obtained into the host cell, culturing the transformed cell in a medium under conditions favorable to the production of proteins, and recovering the medium containing the receptor protein fragment from the cultured cell. The composition containing the protein fragment thus obtained is also provided.
