Abstract:

PURPOSE: To obtain the subject yeast capable of efficiently producing squalene on an industrial scale by carrying out a gene operation by a specific host yeast and a yeast-E.coli shuttle vector, thereby causing the mutation on a steroid metabolic path.

CONSTITUTION: A homologous recombinant genetic operation is carried out by a host yeast having developed host-vector system [preferably Saccharomyces cerevisiae SHY3 (ATCC-44771)] and a yeast-E.coli shuttle vector (preferably plasmid pIAO2) to induce a mutation on a sterol metabolism path (destruction, degeneration and/or elimination of a squalene epoxidase gene) to obtain the objective yeast.
