Abstract:

PURPOSE: To produce the subject cellulase-free enzyme capable of breaking down xylan into cellulose by enzymatic treatment of an unbleached pulp by culturing a fungus Thermomyces langinosus in a nutrient culture medium containing a corn cob.

CONSTITUTION: A preculture of Thermomyces languinosus DSM5826 is inoculated into a nutrient culture medium containing corn cobs and cultured at a temp. of 45-50°C and a pH value of 6.0-7.0 for 4-5 days while shaking and then centrifuged to remove an insoluble substance such as a microbial cell. The culture supernatant is purified by salting out, gel filtration, ion exchange chromatography and gel electrophoresis, etc., to afford the objective exo- and endo-celluase-free xylanase capable of enzymatically treating an unbleached pulp, breaking down a hemicellulose such as xylan an producing cellulose.
