Abstract:

PURPOSE: To provide a DNA fragment expressed by a specific base sequence containing a gene of Pseudomonas strain to produce a lactone-synthesizing lipase.

CONSTITUTION: Pseudomonas nov. sp. No.109 (FERM P-3025) is cultured, the cultured cells are collected, a lysozyme solution is added and the mixture is left standing for a prescribed period. An SDS solution is added to the mixture to effect the complete bacteriolysis and DNA is extracted by treating with phenol. The DNA is recovered by precipitating with isopropanol and treated with an RNase to obtain a chromosomal DNA.
