PURPOSE: To separate and purify the titled substance from a plasma protein mixture liquid, by extracting the specific bonding site of antithrombin III from natural sulfated hydrocarbon, bonding the extract to a hydrophilic carrier, and contacting the resultant gel with the plasma protein mixture.
CONSTITUTION: Heparin which is a natural sulfated mucopolysaccharide, or its acetylated derivative, is decomposed by digesting with a specific enzyme such as heparinase, etc. or by deamination decomposition. The obtained oligosaccharide is purified by affinity chromatography, etc. using antithrombin III sepharose, lysine sepharose, etc. to obtain a fragment bonded with antithrombin III. The ligand is immobilized to a hydrophilic carrier by a firm bonding method, and the obtained gel is brought into contact with a plasma protein mixture liquid to effect the specific adsorption of the antithrombin III from the mixture. The adsorbed antithrombin III is desorbed to obtain purified antithrombin III.
EFFECT: The purification can be carried out rapidly and easily in high efficiency, and the ingredient proteins can be effectively removed by this process.
