PURPOSE: To provide a method useful for amplifying a nucleic acid by using PCR and to provide a diagnostic test kit.
CONSTITUTION: This PCR reagent can be encapsulated with an inert meltable material which will keep the reagent from interaction in unwanted amplification reactions until its reaction is desired in the amplification of a targeted nucleic acid. Various reagents used in PCR can be so encapsulated, but particularly the DNA polymerase coreagents and deoxyribonucleotide-5'-triphosphates. The encapsulating material is a water-impermeable solid at room temperature, but melts at a temperature of at least about 50°C at which primer extension products are generally formed. The encapsulated PCR reagent can be used in various amplification and detection methods to reduce nonspecific amplifications.
