Abstract:

PURPOSE: To enable transduction of an exogenote without applying stress to animals cells by adding a complex of the objective gene to be transduced and lactoferrin into a culture solution of the animal cells, allowing the resultant mixture to stand and incorporating the complex into the cells.

CONSTITUTION: Modified lactoferrin or lactoferrin is dissolved in a previously prepared buffer solution and a prescribed amount of a gene is further added to gently stir and mix both. The resultant solution is then allowed to stand at a prescribed temperature for a prescribed time to produce a DNA lactoferrin complex. A buffer solution containing a prescribed amount of the resultant DNA lactoferrin complex is subsequently added into a culture solution in which cells to be transduced are precultured and culturing is continued for a prescribed time. The DNA sticks through the lactoferrin to cell surfaces and incorporated into the cells. If the gene to be transduced is marked with a selective marker, the cells into which the gene is transduced can readily be selected after completing the culturing. After the transduction of the DNA, the culture solution can be exchanged with a culture medium containing a chemical for selection added thereto to carry out culturing for a prescribed period. Thereby, the objective survival cells as cells in which the gene is transduced are obtained.
