NEW MATERIAL:A human laminine fragment obtained from human body fluid, having a molecular weight of about 42,000 determined by SDS-polyacrylamide gel electrophoresis and containing an N-terminal amino acid sequence of formula (X is unidentified amino acid residue).
USE: A detection reagent for malignant tumor, hepatic diseases, renal diseases and arteriosclerosis.
PREPARATION: The objective human laminine fragment of formula can be produced e.g. by homogenizing human placenta in 0.05M tris-HCl buffer solution (pH7.2) containing 0.5M sodium chloride, removing insoluble materials by centrifugal separation, adding sodium chloride in an amount to attain the final concentration of 4M, collecting the produced precipitate, dissolving the precipitate in a 0.05M tris-HCl buffer solution (pH7.2) containing 0.5M sodium chloride, fractionating the solution by gel-filtration with a column equilibrated with the above buffer solution and collecting the fraction eluted as the first peak.
