Abstract:

PURPOSE: To obtain a large amount of a peptide hormone sample as a purified substance in a short time, by means of liquid chromatography using a specific hydrophobic chromatographic packing.

CONSTITUTION: A hydrophobic chromatographic packing obtained by introducing hydrophobic groups, e.g. 1W10C aliphatic alkyl, phenyl or cyclohexyl groups, into spherical semirigid crosslinked polymer particles (preferably having 10W 150μ average particle diamete and ≥3k/cm2 pressure resistance) having macropore diameter with 3W5ml/g-dry gel water adsorption and ≥105 exclusion limit molecular weight expressed in terms of standard polyethylene oxide is used to subject peptide hormone to liquid chromatography to give a purified peptide hormone. Spherical porous crosslinked polymethacrylate having polyol ether or polyol polyether residue as alcoholic residues is preferred for the above-mentioned crosslinked polymer.
