Abstract:
PURPOSE: To provide a method for respectively glycosylating an acceptor with an N-acetylglucosamine(GlcNAc) or an N-acetylgalactosamine(GalNAc) group while recycling uridine 5'-disphosphate in one pot.
CONSTITUTION: The following components (i) to (iv) are mixed in (a) an aqueous medium in one pot to form (b) an aqueous reaction medium, which is then maintained at a pH value of about 5 to about 10 for a time sufficient to glycosylate the acceptor at a temp. from about 0 to about 40°C to glycosylate the acceptor with GlcNAc or GalNAc. Uridine 5'-diphosphare recycle system containing (i) GalNAc-1-P or GlcNAc-1-P, (ii) a pyrophosphorylase, (iii) a transferase, (iv) an epimerase, (v) the acceptor and (vi) (a) uridine 5'-diphosphate(UDP) or uridne 5'-triphosphate(UTP) or both, (b) a phosphate donor and (c) a kinase capable of transferring the phosphate group from the phosphate donor to the UDP and forming the UTP.
