PURPOSE: To obtain the subject merlin gene of a tumor suppressor gene, encoding merlin protein and useful for early diagnosis of genepathy associated with the merlin gene, detection and treatment of tumor such as neurofibromatosis 2, and gene therapy or the like.
CONSTITUTION: The objective merlin gene useful for detection of a predisposition to easily suffer from tumor such as neurofibromatosis 2(NF2), diagnosis, treatment and monitoring of the treatment of the tumor, a gene therapy such as gene substitution in nonneoplastic tissue or cell having the tumor, or the like is obtained by gathering mRNA from the tumor of a patient suffering from the NF2 by a routine procedure, synthesizing cDNA by using the gathered mRNA as a template, forming a cDNA library, cloning the cDNA library by using an oligonucleotide probe specific to a merlin sequence, gathering the DNA from the positive clone, and treating the gathered DNA with a restrictive enzyme. The gene is incorporated into a vector and the obtained vector is allowed to express in a host to provide the merlin sequence.
