NEW MATERIAL:A compound of the formula (R1 is H, halogen; R2 is H, or incorporates R1 to form single bond; R3 is H, lower alkanoyl).
EXAMPLE: 4'''-Deoxytylosin.
USE: It is a 16-membered macrolide antibiotic and used as an antibacterial. It is stable in living bodies.
PREPARATION: For example, Streptomyces fradie NRRL B-2702 strain is cultured in an enriched medium and the resultant tylosin is slectively acylated only in the 2'-position with a lower alkylcarboxylic acid, its acid halide or anhydride, in the presence of a base to give 2'-O-acyltylosin. The product is allowed to react with a halomethanesulfonic acid anhydride to obtain the corresponding 4'''-O-sulfonylated product. Finally, the sulfonylated compound is treated with a halogenating agent.
