Abstract:
PURPOSE: To obtain a vaccine composition inhibiting growth, etc., of epithelial growth factor-depending tumor without introducing foreign protein by an autoimmune effect, containing an autochthonous epithelial growth factor combined with a carrier protein and an adjuvant.
CONSTITUTION: This vaccine composition comprises (A) an autochthonous epithelial growth factor(EGF) combined with a carrier protein (preferably cholera toxin B-chain, tetanus toxin, monoclonal antibody, etc.) and further contains (B) an adjuvant (preferably aluminum hydroxide). Preferably, a human recombinant epithelial growth factor (hu-recEGF), etc., is used as the autochthonous EGF. The component A is obtained by, e.g. mixing a solution of hu-recEGF in 10 mM PBS/MgCl2 with a solution of cholera toxin B-chain in the same solvent at a ratio of 1-5 mol of EGF per 1 mol of the protein, adding 0.5 % glutaric aldehyde, incubating at a room temperature and dialyzing in 10 mM PBS/MgCl2.
