Abstract:
PURPOSE: To provide a new kind of vitamin D compound suitably obtained from an easily available or accessible starting substance, particularly a vitamin D2 compound and a related compound.
CONSTITUTION: There is provided a vitamin D compound of formula I [wherein R1 is H or an OH protecting group; Z is formula II or III (wherein R2 and R3 are each a 1-6C alkyl or 2-6C alkenyl, optionally substituted with a substituent selected from OH, an alkoxy, F, cycloalkyl and alkylenedioxy or with an optionally substituted phenyl, or 3-6C cycloalkyl or optionally substituted phenyl) or R1 and R3 together with an interconnecting oxadimethylene divalent group form a 5- or 7-membered ring-closed 1-4C alkyl substituted hemiacetal; and A and B are each H or methyl, or form together CH2]. The compound is obtained by reacting a compound of formula IV with a reagent of the formulae: R3-MgX or R3-Li in the presence of a Cu(I) compound, oxidizing a resultant compound of formula V after the removal of the protecting group of OR4 to obtain the corresponding hydriodan-4-one compound, converting with a Wittig reactant of formula VI, and subsequently hydrogenating and isomerizing.
