Abstract:
PROBLEM TO BE SOLVED: To provide chimeric factor VIII polypeptides and uses thereof.SOLUTION: The invention provides a VWF fragment comprising VWF D' domain and D3 domain, a chimeric protein comprising the VWF fragment and a heterologous moiety, or a chimeric protein comprising the VWF fragment and FVIII protein, as well as a method for using them. The polypeptide chain comprising the VWF fragment binds to or associates with a polypeptide chain comprising FVIII protein, and the polypeptide chain comprising the VWF fragment can prevent or inhibit binding of endogenous VWF to FVIII protein. By preventing or inhibiting the binding of endogenous VWF, which is a half-life limiting factor of FVIII, to FVIII, the VWF fragment induces extension of half-life of the FVIII protein. The invention encompasses nucleotides, vectors, host cells, methods for using the VWF fragment or the chimeric proteins.SELECTED DRAWING: None
