Abstract:
PROBLEM TO BE SOLVED: To provide a method for freezing iPS-NSPCs applicable to transplantation therapy for spinal cord injury.SOLUTION: According to the present invention, the method comprises: a cooling step of cooling and freezing a sample consisting of a cell suspension in which a cryopreservation solution is added to human iPS cell-derived neural stem/progenitor cells that form a neurosphere; and a melting step of melting the frozen sample in a container by a rapid melting method. The cooling step uses a refrigerating apparatus 1 including a freezing tank 11, means (16, 22) for generating a magnetic field in the freezing tank, and means 25 for variably controlling the inner temperature of the freezing tank. After placing the container containing the sample in the freezing tank, while applying an alternating magnetic field to the sample, the container is cooled such that the inner temperature of the freezing tank is changed from initial temperature T1 to temperature T2 that is lower than the freezing temperature of the sample, and the inner temperature of the freezing tank is controlled to be kept for a predetermined time at temperature T3 at which the cryopreservation solution is in a supercooled state during the process of reaching temperature T2.SELECTED DRAWING: Figure 1
