PROBLEM TO BE SOLVED: To provide a compound effective for treating: diabetic gastroparesis, and impairment of hypomotility in the gastrointestinal tract such as constipation type irritable bowel syndrome; CNS related disorders such as migraine, schizophrenia, psychosis or depression; and glaucoma and preterm labor related ocular hypertension.SOLUTION: Provided is a 12-30 membered macrocyclic compound, as represented by a compound of example 1 in scheme 6, having the conformation fixed and having agonist or antagonist activity to motilin receptor (MR receptor), 5-HTsubtype of serotonin receptor (5-HTreceptor), and prostaglandin F2A receptor (FP receptor).SELECTED DRAWING: None
