Abstract:
The present invention provides a very safe Ad vector exhibiting a gene transfer efficiency equivalent to or greater than that of conventional Ad vectors; i.e., an Ad vector capable of being effectively and safely used in gene therapy and life sciences research. Further provided are Ad-vector-producing/-amplifying cells capable of amplifying the Ad vector. According to this Ad vector, a VA-RNA gene (VA-RNA I and/or VA-RNA II) is modified in the Ad vector genome so that the VA-RNAs are not expressed. In Ad-vector-producing/-amplifying cells, a gene is introduced so that the VA-RNAs of the Ad genome are expressed in cells constitutively expressing an E1 gene product used to amplify the Ad vector.
