Abstract:
The purpose of the present invention is to provide: a high-quality composition containing alkaline phosphatase; a method for producing dephosphorylated nucleic acid using the composition; and a method for producing labelled nucleic acid using the composition. To achieve the purpose, the present invention provides a composition containing alkaline phosphatase, and a peptide fragment group (A) including two or more peptide fragments each including 5-50 consecutive amino acid residues selected from positions 501-578 of the amino acid sequence of SEQ ID NO: 5, wherein the ratio of the total contained amount of one or more of the peptide fragments with respect to the contained amount of the alkaline phosphatase satisfies formula (A): (XA/Y) × 100 ≤ 4.4000 (in the formula, XA represents the peak area value of the peptide fragment group (A) as calculated by an automatic integral technique from an extracted ion chromatogram obtained by LC-MS/MS analysis on the composition, and Y represents the peak area value of the alkaline phosphatase as calculated by an automatic integral technique from a chromatogram obtained by LC-UV analysis on the composition).
