Abstract:
The present invention provides an anti-CD28 humanized single-domain antibody. Specifically, disclosed are a plurality of single-domain antibodies targeting CD28 and humanized antibodies thereof, derived proteins, gene sequences encoding same, and expression vectors and expression systems for producing same. The single-domain antibody of the present invention is a CD28 non-agonistic antibody, can block binding of CD28 to CD80, and can specifically bind to human and monkey CD28. The single-domain antibody of the present invention has high safety and provides a research and development basis for development of antibody drug conjugates and multispecific antibody drugs. A bispecific antibody formed by the CD28 single-domain antibody of the present invention and an antibody targeting a tumor antigen can remarkably enhance the activity of a CD3 bispecific antibody, and a multispecific antibody formed by the CD28 single-domain antibody of the present invention, the antibody targeting a tumor antigen, and the CD3 antibody has activity remarkably superior to that of the CD3 bispecific antibody.
