Abstract:
An apparatus (50) for preparing samples consisting of a sample preparation unit (100) having an upper opening, into which a buffer having cells suspended therein is poured, a lower opening, from which a waste liquor is discharged, and a channel connecting these openings, provided with a first filter (110) for capturing cells, a second filter (130) for capturing polynucleotides eluted from the cells with a denaturing agent poured into the upper opening and holding a PCR amplification reaction mixture containing a fluorescence-labeled primer for PCR and a PCR amplification reaction reagent for amplifying the copy of a desired partial base sequence in the polynucleotide, and a third hydrophobic filter (140), wherein these filters are arranged in this order from the upper opening toward the lower opening; a member (200) for holding the sample preparation unit; and a member (210) for controlling the temperature of the PCR amplification reaction mixture. By using this apparatus, a series of steps including the pretreatment of specimen samples, PCR amplification and the detection of the PCR product can be continuously performed. Thus, the contamination at each step can be prevented and a number of specimens can be easily treated simultaneously.
