Abstract:
A novel member of the human AMPK family is identified and named ARK5 (AMPK-Related Kinase 5). In nutritional hunger, ARK5 Akt-dependently supports the survival of cells. A P/ARK cell line constructed by stably transfecting PANC-1, which is a human pancreatic cancer cell line, with ARK5 shows an in vitro metastatic activity twice or more as high as PANC-1 does. In a nude mouse, the P/ARK cell line shows more than 10-fold increase in tumor proliferation 12 weeks after transplantation. The P/ARK cell line shows a significant decrease in the necrotic area in a transplanted tumor nest compared with the parent PANC-1 cells. A nude mouse having the P/ARK cell line transplanted thereto shows obvious intramuscular infiltration and mesenteric metastasis.
