Abstract:
A base editing system based on bimolecular deaminase complementation, and use thereof. The base editing system is mainly composed of nCas9 and nucleobase deaminase double complementary base editing fusion proteins A and B, as well as a guide RNA. While the high on-target efficiency in cytosine base editing is maintained, the Cas9-dependent off-targeting and Cas9-independent off-targeting on the genome of the base editing system are greatly reduced. Further provided is an adenine base editing system based on bimolecular deaminase complementation. The cytosine and adenine base editing systems based on bimolecular deaminase complementation are generally suitable for various eukaryotic organisms, and can be used in fields such as crop genetic breeding, animal variety improvement, and even clinical treatment of human genetic diseases.
