Abstract:
A base editing system and method constructed based on a CRISPR/Cpf1 system are provided. The base editing system utilizes Cpf1 to recognize features of a 5'-TTTN-3' (PAM) sequence, and carries out base editing in regions rich in adenine and thymine in genomic DNA. The base editing system comprises at least one of a Cpf1-BE0 expression vector, a Cpf1-BE expression vector, a Cpf1-BE-YE expression vector, a Cpf1-BE-UGI expression vector, an RNA product expressed by the Cpf1-BE0 expression vector, an RNA product expressed by the Cpf1-BE expression vector, an RNA product expressed by the Cpf1-BE-YE expression vector, an RNA product expressed by the Cpf1-BE-UGI expression vector, a protein product expressed by the Cpf1-BE0 expression vector, a protein product expressed by the Cpf1-BE expression vector, a protein product expressed by the Cpf1-BE-YE expression vector, and a protein product expressed by the Cpf1-BE-UGI expression vector. A method for carrying out base editing in the genome of a eukaryote is also provided.
