Abstract:
Disclosed in the present invention are a cell-free DNA-based cancer diagnosis model and the use. In a first aspect of the present application, provided is a construction method of a cancer diagnosis model, the construction method comprising the following steps: acquiring sequencing data of cell-free DNAs of a non-cancer population; comparing the sequencing data of the cell-free DNAs with a reference genome to obtain corresponding sites of the tail ends of the cell-free DNAs on the reference genome; and constructing a cancer diagnosis model according to the corresponding sites of the tail ends of the cell-free DNAs on a target genome range. The present application uses an idea opposite to a mainstream method, and constructs a cancer diagnosis model by means of searching for a tail end distribution mode of cfDNAs of a non-cancer population, instead of attempting to identify a diagnosis marker based on a tumor cfDNA specific fragmentation mode. By means of the manner, patients with a plurality types of different cancers can be distinguished from a contrast population, achieving good specificity and sensitivity, and realizing pancancer type cancer diagnosis.
