Abstract:
Provided is a composition or kit for detecting a target nucleic acid sequence variant. The kit comprises: a closed probe, a detection probe, and a primer; the closed probe is combined with a wild type variant of a target nucleic acid sequence by means of specific binding, and a tail end 3' thereof is modified with oligonucleotide for preventing its extension; the detection probe is combined with a mutant variant of the target nucleic acid sequence by means of specific binding and can generate a detection signal; the primer is a common primer for the wild type variant and the mutant variant of the target nucleic acid sequence. Also provided is a method for detecting a target nucleic acid sequence variant. The method achieves a purpose of high selectivity detection by enriching mutant variants of a target nucleic acid sequence in a nucleic acid sample by means of a PCR reaction, and finally detecting an amplicon according to changes in detection signals of a detection probe.
