Abstract:
The present invention provides construction and application of a novel bispecific chimeric antigen receptor (CAR), and relates to the technical field of immunotherapy. The CAR provided by the present invention comprises an antigen-binding domain, a linker peptide, a hinge region, a transmembrane domain, a 4-1BB costimulatory signaling region, and a CD3ζ signaling domain. The antigen-binding domain comprises an anti-CD19 scFv and an anti-CD22 nano antibody, or comprises an anti-HER2 scFv and a ligand capable of identifying IGF1R. A CAR-T provided by the present invention has the capability of simultaneously binding to two target antigens, and has stronger killer activity and cytokine release capability compared with a single-target CAR-T. A bispecific CAR-T cell provided by the present invention is a bispecific CAR-T cell based on an antiparallel β-stranded loop linker (BS Loop), and the two antigen-binding domains can both play a good role, without affecting each other, and have stronger tumor killer activity and T cell activation and proliferation capacity compared with other linear tandem bispecific CAR-T cells or reported bispecific CAR-T cells of the BS Loop.
