Abstract:
A process for producing curculin B, which comprises culturing a transformed microorganism or cell containing inherently pure curculin B or a recombinant DNA containing a base sequence which codes for curculin B to thereby produce curculin B and isolating the produced curculin B from the transformed microorganism or cell. A process for producing a DNA containing a base sequence coding for curculin B, which comprises isolating a fraction containing mRNA of curculin B from a DNA containing a base sequence coding for curculin B or its precursor or from Curculigo latifolia, preparing a single-stranded cDNA from the mRNA with a reverse transcriptase, preparing a double-stranded cDNA from the single-stranded cDNA, inserting the double-stranded cDNA into a vector, transforming the host with the resultant vector to prepare a cDNA library, and isolating a cDNA coding for curculin B from this library by using a probe comprising a synthetic DNA containing a base sequence coding for a partial amino acid sequence obtained by analyzing purified curculin A obtained from Curculigo latifolia.
