Abstract:
A genomic DNA amplification method, comprising: (a) providing a first reaction mixture comprising a sample containing genomic DNA, a first primer, a nucleotide monomer mixture, and nucleic acid polymerase, the first primer comprising, from a terminal 5' to a terminal 3', a universal sequence and a first variable sequence comprising a first random sequence; (b) applying the first reaction mixture to a first temperature cyclic procedure, to obtain a product to be amplified; (c) providing a second reaction mixture comprising the product to be amplified, a second primer, a nucleotide monomer mixture, and nucleic acid polymerase, the second primer, from a terminal 5' to a terminal 3', comprising or being composed of a particular sequence and the universal sequence; and (d) applying the second reaction mixture to a second temperature cyclic procedure for amplification, to obtain an amplified product. The present invention also relates to a kit for amplifying a genomic DNA.
