Abstract:
A preparation method for a DNA origami-based tumor neoantigen delivery system, comprising: selecting eight single-stranded DNAs, wherein 20 nt ployT structures extend from the 5' ends of three of the single-stranded DNAs and are used for pairing with tumor neoantigen peptide-nucleic acid reversible covalent coupling molecules containing sulfonium salt centers to realize the assembly of an OVA antigen and DNA origami; selecting another three single-stranded DNAs and extending CpG sequences at the 5' ends of the three single-stranded DNAs as immune activation co-factors to cooperate with an immune system for activation; mixing the single-stranded DNAs for preparing the DNA origami and the neoantigen peptide-nucleic acid reversible covalent coupling molecules; and carrying out high-temperature denaturation and annealing hybridization to form a triangular pyramid-shaped DNA tetrahedral nanocomposite, namely a DNA origami-based tumor neoantigen delivery system. Experiments prove that the constructed DNA origami-based delivery system containing sulfonium salt centers significantly inhibits tumor growth and lung metastasis of tumor cells.
