Abstract:
Disclosed is an expression vector which is effective for efficiently establishing a transformed cell capable of expressing a gene for a protein of interest at a high level. Specifically disclosed is an expression vector which comprises: an expression cassette for a drug selection marker gene, which comprises an mRNA-destabilizing sequence; at least one gene expression stabilizing element; and an expression cassette for a gene for a protein of interest. Preferably, the mRNA-destabilizing sequence is derived from an AT-rich sequence occurring in a cytokine, an interleukin or a 3'-untranslated region of a protooncogene, and the gene expression stabilizing element is derived from the genome of a Chinese hamster.
