Abstract:
Provided is a fluorescent probe, containing a) a response polypeptide and b) an optical active polypeptide, wherein the optical active polypeptide is inserted into the response polypeptide. Also provided is a nucleic acid sequence coding the fluorescent probe of any embodiment of the present invention or a complementary sequence thereof. Also provided is an expression vector containing the nucleic acid sequence of the present invention, which is operably linked to an expression control sequence, or the complementary sequence thereof. Also provided is a cell containing the expression vector of the present invention. Also provided is a method for preparing the fluorescent probe of the present invention, the method comprising the steps of: providing the cell containing the vector for expressing the fluorescent probe of the present invention; culturing the cell under the cell expression conditions; and separating the fluorescent probe. Also provided is a use of the fluorescent probe of the present invention or the fluorescent probe prepared by the method of the present invention in detecting branched chain amino acids. In one embodiment, the branched chain amino acids are selected from leucine, isoleucine and valine. The detection can be performed in vivo, in vitro, at subcellular level or at in-situ level. Further provided is a kit containing the fluorescent probe of the present invention or the fluorescent probe prepared by the method of the present invention.
