Abstract:
Provided is a gene mutation introduction method comprising: a step for expressing a fusion protein between a sequence-specific DNA cleavage enzyme and a nuclear receptor under control of an expression-inducing promoter; a step for forming a double-strand DNA break by causing the fusion protein to move into the nucleus and letting the fusion protein that has moved into the nucleus cut genome DNA in a sequence-specific manner; and a step for, by utilizing a DNA break repair mechanism, repairing the double-strand DNA break and introducing a gene mutation into the vicinity of the double-strand DNA break.
