Abstract:
A genetically engineered bacterium and a production method for producing lactyl-N-neotetraose, relating to the technical field of metabolic engineering and food biology. In order to solve the existing problem of low yield of producing lactyl-N-neotetraose by using a microbial method, by means of exogenous expression of lgtA and lgtB, overexpression of lacY, pgm, gale, galT, and galK in a lactyl-N-neotetraose synthesis pathway is reasonably combined and regulated and controlled, a lacZ expression in Escherichia coli hosts is knocked out, and a carbon source configuration in the culture process is optimized, thereby achieving the purpose of regulating and controlling the carbon flux of a metabolic pathway and improving the yield of the lactyl-N-neotetraose. In shake flask experiments, the capability for Escherichia coli to produce lactyl-N-neotetraose is increased from 304 mg/L to 1031 mg/L.
