Abstract:
Provided are a group of isotope labelling reagents for carboxyl labelling and a synthesis method therefor, wherein one end of the isotope labelling reagent is a primary amine and the other end is a trimethyl ammonium salt; and the primary amine and the trimethyl ammonium salt are connected by means of a plurality of carbon chains. The synthesis process involves: protecting primary amino groups, with N,N-dimethylethylenediamine and homologues thereof as reaction raw materials, by using di-tert-butyl dicarbonate ((BOC)2O), and then reacting same with a methyl iodide reagent which contains isotope D and 13C under alkaline conditions, and finally removing the protective group under acidic conditions to obtain a target product. By means of a nucleophilic substitution reaction, a group of isotope labelling reagents which can be used for the high-efficiency labelling of carboxyl groups are synthesised; the price is low, the operation is simple, and after labelling with the isotope reagent, the efficiency and accuracy of analysis can be significantly improved.
