Abstract:
Provided are: a HasA-redoxin complex (a heme iron active-type HasA) which can asymmetrically oxidize only one of enantiomers of a secondary alcohol without the need of adding a coenzyme and has an asymmetric oxidization activity in an aqueous solvent in the presence of oxygen, and an immobilized form of the HasA-redoxin complex; methods respectively for producing the HasA-redoxin complex and the immobilized form of the HasA-redoxin complex; and a production method relating to the coating of the protein complex with a polymeric compound. Methods respectively for producing a HasA-redoxin complex (a heme iron active-type HasA) having an asymmetric oxidation reaction and an immobilized form thereof. The methods are characterized by comprising: a first step comprising a treatment for encapsulating a plant-originated water-soluble crude protein in a gel, a treatment for oxidizing the gel with air, and a treatment for eluting a HasA-redoxin complex from the gel into an aqueous solution; and a second step comprising a treatment for applying a gravity to the aqueous HasA-redoxin complex solution to concentrate and precipitate the HasA-redoxin complex, a treatment for re-dissolving the HasA-redoxin complex precipitates in an aqueous solution to allow the HasA-redoxin complex and a polymeric compound to co-exist homogeneously, and dehydration/drying of the co-existing aqueous solution to produce a polymeric compound-coated HasA-redoxin complex. The methods are also characterized in that HasA is allowed to express using a transformant (a microorganism) containing HasA originated from Pseudomonas fluorescens, and then the HasA is agitated and oxidized with air in an oxygen-containing aqueous solution, thereby producing a HasA-redoxin complex (a heme iron active-type HasA).
