Abstract:
A high-throughput body fluid protein sample pretreatment apparatus. The apparatus is a system integrating high-abundance protein removal from body fluids (plasma, serum, and urine), and medium- and low-abundance protein online denaturation and reduction, desalting, and on-line enzymolysis, and comprises: a high-abundance body fluid protein removal antibody column (3), a cluster type high temperature denaturation and reduction device, a cluster-type solvent displacer, and an immobilized enzyme reactor (12). High-abundance proteins are first removed from a body fluid protein sample by means of the antibody column (3); then the eluted medium- and low-abundance proteins are then subjected to online and rapid denaturation and reduction by the cluster-type high-temperature denaturation and reduction device; the denaturing agent and the reducing agent are removed by means of the cluster-type solvent displacer; finally, online enzymolysiss is conducted by the immobilized enzyme reactor (12), and peptides generated by means of enzymolysiss can be tested by means of mass spectrometry. The high-throughput body fluid protein sample pretreatment apparatus integrates complex sample pretreatment processes for body fluid proteins.
