Abstract:
Provided are a high-throughput multiparametric single cell-derived extracellular vesicle analysis chip and use thereof. The chip comprises a two-layer structure, one layer being a bioanalytical substrate for extracellular vesicle capture and detection, the other layer being a chip with a high throughput array for single cell capture. The microfluidic chip technology and the single cell analysis technology are applied to the study of extracellular vesicles, and high-throughput and multiparametric capture can be achieved by using an immunoaffinity method, thereby detecting extracellular vesicles of various sizes secreted by a single cell. Further, qualitative and quantitative detections, correlation analysis, multifunctional analysis, and immunophenotyping analysis of the secretion of extracellular vesicles at a single cell level are carried out, clearly demonstrating the heterogeneity of cells and the secreted extracellular vesicles among the cells.
