Abstract:
Provided is an autoinduction expression system in which variations are applied to Acs (acetyl-CoA synthase) gene promoters (mutagenesis) and an Acs promoter library is built up, and which shows more diverse expression aspects (expression volume and expression profile) than natural-occurring Acs promoters. The autoinduction expression of the present invention takes place with adjustment of the intracellular network for sensing the energy and nutritional states. In contrast to induction methods of the prior art which necessitate the monitoring of respective culture growths and the addition of inducers (IPTG or lactose) at appropriate times, the autoinduction expression system of the present invention can be implemented very simply, and allows target proteins to be obtained in large volume or with diverse profiles when cells which have been genetically transformed using an autoinduction expression vector of the present invention are inoculated into a culture medium having a carbon source (e.g. glucose) and culturing is carried out up until the stationary phase is reached.
