Abstract:
Disclosed is a method and device for on-line monitoring the process of protein enzymatic hydrolysis based on in-situ real-time spectrum, which belongs to the field of on-line monitoring of protein enzymatic hydrolysis process. Enzymatic hydrolysis is made to gluten protein suspensions of different concentrations, timing sampling is made during the enzymatic hydrolysis process, and important parameters throughout the enzymatic hydrolysis process are monitored using chemical methods, wherein these parameters include: degree of hydrolysis, concentration of polypeptides in enzymatic hydrolysate, and ACE inhibition rate of the enzymatic hydrolysate. The in-situ real-time spectrum of the collected enzymatic hydrolysate is quickly collected. The collected spectrum is pretreated. The screening of the optimum spectral intervals for the degree of hydrolysis, the concentration of polypeptides in the enzymatic hydrolysate, and the ACE inhibition rate of the enzymatic hydrolysate are made by using a joint interval least square method. A calibration model and a prediction model are established by using the joint interval least square method. And the in-situ real-time monitoring and the end of the reaction are determined with respect to the enzymatic hydrolysis process using the above-mentioned prediction model. The enzymatic hydrolysis process of gluten protein suspension with substrate concentration of 10 g/L is monitored by the prediction model, so that the coincidence degree between the predicted value and the measured value is relatively high.
