Abstract:
A method of in vitro polymerizing a heparan chain with the use of a polymerization initiator having a structure containing at least trisaccharide GlcNAc&agr 1-4GlcUAβ1-3Galβ or disaccharide GlcUAβ1-3Galβ among pentasaccharides within GAG-protein binding region, which is bound to a hydrophobic aglycone. In the polymerization reaction, it is preferred to use a product of co-expression of secretory EXT1 and EXT2 as an enzyme. However, in the use of specified polymerization initiator, a heparan chain can be synthesized even with the use of secretory EXT1 alone.
