Abstract:
Provided is an intein C-terminal sequence variant, wherein compared with an amino acid sequence as shown in SEQ ID NO. 1, the intein C-terminal sequence variant contains at least one mutation positioned at amino acid positions selected from a group consisting of K2, K7, K11 and K28. The intein C-terminal sequence variant can be used to participate in the preparation of antibody-conjugated drugs, including antibody-drug conjugates, antibody-supramolecular nanofiber conjugates (such as photothermal therapy ICG, etc.) and antibody-protein degrading agent conjugates (such as PROTAC molecules), so that the drugs are subjected to site-specific conjugation. Further provided is a method for preparing a conjugate using the intein C-terminal sequence variant.
