Abstract:
This light emission observation method comprises the steps of: maintaining a biological sample in a viable state, the biological sample containing an observation cell expressing a gene for a first photoprotein; preparing a reference that displays a reference amount of the first photoprotein so that when the observation cell is photographed, the reference is included in one field of view with the observation cell; applying a predetermined stimulation to the observation cell and the reference to cause a luminescent reaction in the observation cell and the reference; and acquiring a plurality of wavelength band images by photographing light emitted from the observation cell and the reference in mutually different wavelength bands. The light emission observation method further comprises steps of: determining, from the plurality of the wavelength band images, the amount of light emitted from the observation cell and the amount of light emitted from the reference; calculating a correction coefficient using the determined amount of light emitted from the reference; and correcting the determined amount of light emitted from the observation cell using the correction coefficient.
