Abstract:
The present invention addresses the problem of developing a technology that can be widely used in mammals but does not require the use of ES cells, wherein the technology (genome editing technology such as ZFN) is for modifying a specific gene using a specific sequence on a genome as the target, and the technology can be used with nothing more than a very simple method. Provided is a technology for modifying with good efficiency any target mammalian gene by immersing a fertilized mammalian egg that has a clear zone and is in the pronuclear stage in a solution containing pair molecules of mRNA having a specific sequence, and performing electroporation by applying three-step square wave multipulses, with the total amount of electricity of the first electrical pulse set to be within a predetermined range.
