Abstract:
Application: for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and besides in cosmetic procedures. Goal: The expansion of medical preparations assortment. The essence of the invention: the preparations wherein the required solutions is recovered at autotrophic iron oxidizing bacteria cultivation, for example, Th. ferrooxidans on corresponding synthetic media or pulps, containing in a form of a solid materials with sulfide minerals or minerals themselves are an active origin. As a result of bacteria growth and development in such media ions of heavy metals and/or metalloids, having toxic effect on pathogenic micro flora are accumulated in the solutions the oxygenation process in the pathological zone, promoting the acceleration of detachment of necrotic masses and damaged tissues reparation is considerably improved.

Claims:
CLAIMS
1. The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic procedures, wherein a solution, containing ions (III) and/or vital cells of autothropic iron oxidizing bacteria, for example Th. ferrooxidans and/or extra cellular metabolites of these bacteria and/or extract thereof is an active origin.
2. The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic purposes wherein, as set forth in claim 1 the required solution is recovered as a result of autothrophic iron oxidizing bacteria growth and development on standard synthetic media, used for these bacteria cultivation or on modifications thereof.

The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and anima) s treatment and prophylaxis and for cosmetic purposes, wherein as set forth in claim 1 the required solution is recovered at autotrophic iron oxidizing bacteria growth and development in pulps, containing in a form of a send sulfide minerals, for example pyrite, chalcopyrite, arsenopyrite and/or ores, and/or concentrates with these minerals, the solution is separated from the solid by filtration during and/or at the end of the cultivation.

The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic purposes, wherein as set forth in claim 1 the soluble compositions of another metals, for example copper and/or silver and/or zinc, having bactericidal and/or bio stimulating effect in concentrations not causing the toxic effect on normal cells is entered additionally into the recovered solution.

The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic purposes, wherein as set forth in claim 1 the soluble compositions of metalloids, for example, arsenic, having bactericidal and/or bio stimulating effect in the concentrations not causing the toxic effect on normal cells are additionally added into the recovered solution.
6. The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic purposes, wherein as set forth in claim 1 the recovered solution is used as applications.

The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic purposes, wherein as set forth in claim 1 the recovered solution is used in the compositions with pharmaceutical applicable thickening agent, for example glycerin.
8. The preparation for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors at human beings and animals treatment and prophylaxis and for cosmetic purposes, wherein as set forth in claim 1 the recovered solution is used for oral cavity rinsing (at stomatitis, paradanthosis etc.).
Description:
MEDICAL-COSMETIC PREPARATIONS FOR SKIN, MUCOUS MEMBRANES From the ancient times the bactericidal action of noble metals ions (silver, gold) is known. The action effect of noble and heavy metals ions in the treatment preparations is not used in practice.

The analog to the proposed preparations is unknown for authors on the basis whereof the authors consider the preparation as a new one.

The invention relates to the field of medicine and can be used for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors treatment and prophylaxis at human beings and animals, besides it can be used in cosmetic procedures.

The goal of invention is the expansion of the medical preparations assortment for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors treatment and prophylaxis at human beings and animals, besides it can be used in cosmetic procedures.

The preparations (solutions) are the product of the vital activity of Thiobacillus ferrooxidans autotrophic bacterium (absolutely non pathogenic for human beings and animals).

The treatment action effect at surface application is connected with the following: 1) high content of iron ions (III) in the solution due whereof the following is observed: a) clearly defined bactericidal and anti fungous effect; b) effect on skin leading to the tanning effect, detachment of necrotic masses, wounds cleaning, wound accelerated healing stimulation; c) inflammatory points trophics increase; 2) with the reduced pH value of 2,5-3, 0 solution destructively affecting the suppressing amount of bacteria and fungi species;

3) with extra cellular metabolites of the Thiobacillus ferrooxidans cells themselves are presented by the broad spectrum of various combinations: organic acids, peptides, polypeptides, aliphatic acids etc ;'the set of these combinations stimulates the wounds healing on skin integument, healing of ulcers, cuts, burns.

The preparations is effective both at application immediately after the damage and later in particular at necrotic processes; 4) besides it is known that high dispersed iron powders (Patent RU N° 2123329, published 12.20. 1998) are used as a anti tumor preparation. From the author's point of view the availability of Fe ions in the applied solution will give the same effect.

The given goal is solved by the way wherein the solutions, recovered at the cultivation of autotrophic, iron oxidizing bacteria, for example Thiobacillus ferrooxidans BKM-458 on corresponding synthetic media or pulps, containing solid materials with sulfide minerals or minerals themselves are used for skin and mucous membranes inflammatory diseases, ulcers, wounds, burns, tumors treatment and prophylaxis and can be used in cosmetic procedures. As a result of bacteria growth and development in such media, ions of heavy metals and/or metalloids, having toxic effect of pathogenic micro flora are accumulated in the solutions. Besides, the cells themselves and their extra cellular metabolites, in particular aliphatic acids have the stimulating effect and/or bactericidal effect.

The invention is illustrated by the following examples: Example 1 Th. ferrooxidans bacteria strain BKM-458 cultivation is carried out on synthetic medium 9K of the following composition: FeS04 7H20-44, 4 g/l, (NH4) 2S04-3 g/l, K2HP04-0, 5 g/1, Mg04-0, 5 g/1, CaCl2-0, 01 g/l

pH of the original medium is approximately 2, 0 and is achieved by the introduction of sulphuric acid (H2SO4) into the solution.

The prepared medium is inoculated by the bacteria culture at the ratio of 1: 100 (10 ml of culture is entered per 1L of medium) and bacteria cultivation is carried out in an incubator at the constant aeration and temperature of 28-30°C up to the iron oxidation level of approximately 80% (80% of Fe (III) of the total amount of the iron). The cultivation time is 3 (three) days. The recovered solution is used as the applied preparation.

Example 2 The same, but the amount of salts is decreased 4 times. 9K 1/4 is the modified medium.

Example 3 Th. ferrooxidans bacteria cultivation is carried out in the medium, containing the copper concentrate as a solid one. 100 g of the concentrate, containing 20% of copper, mainly (90%) as a chalcopyrite, are poured by 11 of fresh water, wherein 3 g of (NH4) 2SO4 and 0,5 g of K2HP04 are dissolved.

The recovered pulp is acidified by sulphuric acid with 1,84 density in the amount of 1,5 ml and inoculated by 100 ml of Th. ferrooxidans cells culture, adapted to the growth and development in these conditions. The cultivation is carried out with constant air aeration, which saturates the solution by oxygen and mixes the solid compound, and temperature 28-30°C in the incubator. As a result the copper and iron (III) pass into the solution. In 10 days the solution is separated from the solid by filtration and used as the applied preparation.

Example 4.

The same, but pyrite is used. Iron (III) passes into the solution and sharp acidification up to pH approximately 1,2 takes place. That is why after the solid separation, the correction of pH up to 2,0-2, 5 values by NH40H solution is carried out.

The preparations, proposed by authors considerably accelerate the reparation process of after operation wounds, particularly at middle-aged

people, at expressed reduction of the immune state of the organism, blood circulation breakdowns in the pathological zones as well as considerably accelerates the wounds healing, formed as a result of thermal effect (burns, freezing) besides it accelerates the healing of trophic ulcers. It efficiently effect at skin fungous diseases treatment and prophylaxis.

Bactericidal activity of preparations being applied was estimated by the method of direct action of solutions on living bacterial cells, preliminary cultivated in Petri dishes.

The following test-cultures were used: Salmonella typhi murium Salmonella typhi Shigella sonnei Pseudomonas aeruginosa St. aureus Proteus mirabilis Enterobacter aerogenes Escherichia coli Sterile zones were formed in all variants at the place of preparations actions on the test-cultures.

The authors propose to use the preparations as themselves in a form of the recovered solution applications as well as in composition with pharmaceutical applicable thickening agents, for example glycerin.
