Abstract:
Calculation of the dissociation constant of a molecule interacting with a molecule to be analyzed and immobilized on a metal surface in surface plasmon resonance is realized by an analytic method providing highly reliable measurement results in which the noise level (the noise width of a reference chip) and the baseline variation (the signal variation of the reference chip) are low. In a method for measuring a signal variation of surface plasmon resonance using surface plasmon resonance measuring apparatus comprising a channel system including a cell formed on a metal film and a photodetecting means for detecting the state of surface plasmon resonance by measuring the intensity of a light beam totally reflected from the surface of the metal film in a state where the liquid flow is stopped after the liquid in the channel system is replaced, the dissociation constant of a molecule interacting with a molecule to be analyzed and immobilized on the metal surface is calculated from the results of the measurement of signal variation.
